The article describes methods using liquid chromatography techniques for the independent detection of piperazine designer drugs in biological and non-biological matrices. The benefit of the LC-MS method is the high sensitivity of determinations, while the LC-DAD method ensures high reproducibility of results. The LC-MS method also confirmed the reproducibility of the main fragmentation patterns for the tested compounds. The addition of deuterated analogues as internal standards to the tested samples ensured reproductible quantification.

There are several routes to the synthesis of mCPP, the most common of which is the reaction of diethanolamine with m-chloroaniline. Other methods involve the reaction of m-chloroaniline with bis(2-chloroethyl)amine or the reaction of piperazine with m-dichlorobenzene. The other two isomers of CPP could be made in a similar way.It is unlikely that the mCPP found in illicit products has been synthesised in clandestine laboratories since it is available commercially as the base or as the hydrochloride salt. 5 LC-MS Analysis—Preparation Of Calibration Samples
A new ‘family” of drugs, the piperazines, has emerged on the European drug market as new designer drugs for the last few years. Piperazines are capable of disrupting a person’s ability to think, communicate, and act sensibly. As with other mind-altering substances, use of BZP or TFMPP may jeopardize work or school performance, ruin relationships, and increase the likelihood of involvements in accidents.

These include among others 1-(3-chlorophenyl) piperazine (mCPP), 1-(3-trifluoromethylphenyl) piperazines (TFMPP), 1-benzyl-4-methylpiperazine (MBZP), 1-(4-fluorophenyl) piperazines (pFPP) and 1-cyclohexyl-4-(1,2-diphenylethyl) piperazine (MT-45). Difficulty in assessing the results obtained can be due to the fact that the metabolic processes of related therapeutic drugs may result in the detection of 1-aryl-piperazines in the biological matrices 86. Piperazine derivatives, detected as metabolic products, account for about 10% of the applied dose. The great advantage of the proposed methods is that they can be used to monitor 1-aryl-piperazines as metabolites of therapeutic drugs.

In recent years, the number of new psychoactive substances (NPS) appearing on the illicit drug market strongly increased. Therefore, we determined the most frequently occurring NPS in The Netherlands and combined this with data regarding drug-related intoxications. Data from the Drugs Information and Monitoring System (DIMS) and the Dutch Poisons Information Centre (DPIC) were combined and jointly analyzed. The number of drug samples submitted to DIMS for analysis containing NPS increased from 22 in 2007 to 431 samples in 2013. The most frequently submitted NPS in 2013 included 4-bromo-2,5-dimethoxyphenethylamine (2C-B), 4-fluoroamphetamine (4-FA), methoxetamine (MXE) and 6-(2-aminopropyl)benzofuran (6-APB).
